Establishment of a cell line with doubly-stable expression. Total RNA was extracted from the selected hygromycin-resistant clones in the absence of doxycycline. Two pairs of primers were used to probe the specific neomycin (NeoΓ) and p53 sequences in the tested cells. Lane 1:100 bp marker; Lane 2: HepG2 cells (carrying the wt p53 gene); Lane 3: Hep3B cells (deletion of p53 gene); Lanes 4, 5, and 6: the tested 13T, 15T and 23T clones.
