SEMA5A-induced Met phosphorylation and VEGF-C expression are dependent upon plexin-B3. a HeLa cells were transduced with lentiviruses expressing plexin-B3-specific shRNA or scrambled shRNA (Control Vector). Western blot analysis of Met phosphorylation was performed on shRNA-transfected cells stimulated with 100 ng/mL SEMA5A for 24 h. Plexin-B3 knockdown reduced SEMA5A-induced Met phosphorylation. The bands were quantified and data are representative of at least three independent experiments. Columns, means of three experiments; bars, SD; *P < 0.05 vs. the respective controls. b RT-PCR and c western blot analysis of VEGF-C expression were performed on plexin-B3 shRNA and control shRNA-transfected HeLa cells. Plexin-B knockdown reduced VEGF-C mRNA and protein expression. RT-PCR, reverse transcription-polymerase chain reaction; SEMA5A, semaphorin 5A; shRNA, short hairpin RNA; VEGF-C, vascular endothelial growth factor-C
