CDDP alters p-EGFR, P-p53 and cleaved-PARP levels and induces ROS-mediated apoptosis in chemosensitive A549 cells but not in chemoresistant H1299 cells. A549 and H1299 cells were cultured with different concentrations of CDDP (0, 2.5, 5 and 10 µM; DMSO as vehicle control) for 24 h. a Protein levels of full length PARP (110 kD), cleaved PARP (89 kD) and GAPDH (loading control) were detected by Western blot. Apoptosis was assessed by Hoechst staining. b Protein contents of P-EGFR (Try 1068), P-p53 (Ser 15), and GAPDH (loading control) were measured by Western blot. c A549 cells were pre-treated with 0, 5 and 10 mM of the reactive oxygen species (ROS) inhibitor N-acetyl-l-cysteine (NAC) for 24 h and treated with CDDP (10 µM, 24 h) followed by Hoechst staining. Quantification and statistical analysis of P-EGFR, P-53 (Ser 15) and, cleaved PARP were conducted. All data were presented as mean ± SEM (n = 4). Data were analyzed by two-way ANOVA followed by Bonferroni post hoc test (*P < 0.05, ***P < 0.001; CTL vs. CDDP)
