TRAF6 mediates signaling transduction pathway. TRAF6 participates in multiple signaling pathways. In the MyD88-dependent pathway of TLR4 signaling pathway, TRAF6 interacts with IRAK and forms the IRAK-TRAF6-TAK1-TAB 1TAB 2 complex and subsequently activates the MAPKs (ERK1, ERK2, p38, and JNK). Ultimately, NF-κB and AP-1 are activated. In the MyD88-independent pathway, TRIF recruits TRAF6 and activates TAK1 to induce NF-κB activation. In RANK signaling pathway, activated RANK interacts with TRAF6 and thereby mediates activation of PI3K-AKT and NF-κB. Akt can phosphorylate Bcl-xL/Bcl-2 associated death promoter (Bad) and Forkhead family members, thus inhibiting apoptosis.
