TLR9 signaling induced the expression of HuR through Akt pathway. 95D cells were treated with 50 μM PI3K inhibitor (LY294002) or specific MEK (Mitogen-activated protein kinase) inhibitor (U0126), 2.5 μM Akt inhibitor IV respectively, and then cultured in 24-well plate in the presence of 10 ug/ml CpG ODNs. 72 hrs later, the expression of HuR protein was determined by western blot and calculated (A).
