PG inhibits autophagy in CCA cells. A HuCCT-1 and TFK-1 cells were treated with different concentrations of PG for 24 h, and the expression levels of LC3B-II/I and P62 were compared by western blot analysis. B TEM was used to analyze the number of autolysosomes in HuCCT-1 and TFK-1 cells treated with DMSO (< 0.1%) or PG (600 nM) for 24 h. A magnified view of the electron photomicrograph shows characteristic autolysosomes. Arrowhead, autolysosomes; N, nuclear. C TFK-1 cells were transfected with GFP-mRFP-LC3B for 24 h and then incubated with DMSO (< 0.1%) or PG (600 nM) for 24 h. Autophagy flux was observed by confocal microscopy. Scale bar: 20 mm. All data are representative of three independent experiments; *P < 0.05
