Impact of PSCs on glycolysis and GEM sensitivity in PCCs. PSC-secreted factors promote glycolysis and induce loss of GEM sensitivity through increased ERK phosphorylation. Glycolysis in PCCs is mediated by increased expression of LDHA and MCT4. Exposure to inhibitors of glycolysis (NV-5440) or MEK/ERK (PD98059) showed protection of PCCs from both PSC-induced loss of GEM sensitivity and enhanced glycolysis as indicated by the increased secretion of lactate.
