BHB attenuates the growth and metastasis of LNCaP tumors transplanted in xenografted nude mice. A, Growth curves of subcutaneous xenografts of LNCaP cells. Nude mice bearing xenograft tumors were treated with BHB (100 mg/kg) or p300 inhibitor A485 (30 mg/kg) for 25 days. The measured tumor volumes versus time were plotted. B, The body weights of mice in various treatment groups were detected. C, Weight of tumors dissected on day 25 after treatment or no treatment. Representative tumor images and statistical charts are shown. D, H&E staining and immunohistochemistry for Ki-67 antigen was performed in xenograft tumors from mice (40×). Scale bars, 100 μm. E, Incidence of metastasis in the bone of mice after BHB treatment or no treatment. Representative images of mouse imaging in vivo and H&E staining in different treatment groups are shown. F, Western blot analysis of CSCs and EMT-associated markers in xenograft tumors from mice on day 25. Data were expressed as the mean ± standard deviation (SD). Paired and two-tailed Student’s t-tests were performed, and significant differences were designated as *p < 0.05, **p < 0.01, and ***p < 0.001
